Production Method
Take 4kg of white willow bark, cut it into small pieces 2 to 5 cm in length, add 15 times the amount of water, and cook
Extract twice, each time 1． 5 h, the two liquids were combined, and filtered, the filtrate was allowed to stand overnight, and the upper clarified extract A (about 95 L) was taken for later use.
Add the above extract A to the macroporous adsorption resin for loading and adsorption (rate: 12 L/h) for more than 24 h, wash with water until the effluent turns light yellow, wash the resin with 20% ethanol to detect salicin by TLC, collect 20% ethanol eluent, and recover ethanol under reduced pressure until there is no alcohol odor to obtain concentrated solution B (about 11 L), which is stored for future use.
Take the concentrated solution B, add n-butanol to extract 3 times, 7 L each time, combine the extracts, and recover n-butanol under reduced pressure. After the recovery is completed, add a small amount of methanol to the sample, continue to recover the solvent, and obtain the crude salicin sample C (about 142.6 g).
Take silica gel (200-300 mesh) 1． 5 kg, add 3 L of chloroform-methanol = 8:1 solution, mix evenly, pack into a column (7.5 cm × 110 cm); take crude sample C, add methanol to dissolve, add 200 g of silica gel and mix the sample (100-200 cm) The ratio is a sample: silica gel = 1: 2), after the solvent is recovered by rotary evaporation, the sample is loaded, and the chloroform-methanol ratio is successively 8: 1, 5: 1, 4: 1 solution and pure methanol gradient washing. The fractions were collected in sections (about 25 L of eluent for each ratio, and 5 L collected in each section). The eluted products were detected by TLC respectively, and the eluates with salicin with little impurity interference were combined, and an appropriate amount of solvent was recovered. After the crystals were precipitated, rinsed with a small amount of methanol and filtered to obtain the crude salicin D (9.5 g), spare.
Take the crude salicin D, add an appropriate amount of methanol (about 500 mL) to heat to dissolve, cool, and crystallize at room temperature, after the crystals are precipitated, filter, collect the crystals, and repeat the operation until the filtrate is colorless, collect the crystals, and dry in a vacuum drying oven to obtain Salicin standard.
