What is Betalain Pigment in Beetroot?

Betalain Pigment in Beetroot is a kind of water-soluble pigment. Like other pigments, it is synthesized in the cytoplasm and stored in the vacuole. It can show rich colors such as red, purple, yellow, and white on plants. At present, the known beet pigments are all quaternary amine alkaloids, which are divided into two types: betaxanthin (Betaxanthin) with an amino acid as prosthetic group and betacyanin (Betacyanin) with glycosylation type. The absorption spectra of betalain and betaaxanthin were 532-550 nm and 457-485 nm, respectively. 


Betaine is the most stable at pH 4-6 and 4 ℃. Therefore, beet root powder extract is widely used in low-temperature refrigerated foods, such as dairy products. The stability of beet pigments to light and temperature is not outstanding enough, but short-term high temperature helps to improve the stability of beet pigments.

What are the characteristics of beet root powder extract?

Betaine pigment in plants not only make flowers beautiful to attract pollinators to progeny. It is also an important factor in realizing the economic value of its flowers and fruits. Physiologically, its presence also plays an important role in plant resistance to stress such as drought stress and peroxidative stress.

The prickly pear, Opuntia ficus, grows in arid regions. It can change the amino acid concentration through the synthesis or degradation of beta-flavin to perform osmotic regulation and improve drought resistance. As an important plant secondary metabolite, beet root powder extract also have antioxidant and photoprotective effects like anthocyanins. Its antioxidant and anti-radiation effects are more pronounced than the membrane protector Trolox (vitamin E analog).


 

How is Betalain Pigment in Beetroot made?

Research on beeline anabolism began in the 1990s. The synthetic pathway mainly includes enzymatic reaction and spontaneous reaction, and the reaction starts from tyrosine. The main enzymes involved in the synthesis pathway are Tyrosinase, UDP-glucosyltransferase, 4,5-DOPA-estradiol-dioxygenase, and Cytochrome P450-like. The first substrate for beet pigment synthesis is L-tyrosine, and tyrosinase has a dual role, and the mycophenolate action of this enzyme produces dopa (L-DOPA). Another heavy activity, Diphenolase, results in the oxidation of dopa to dopaquinone (o-DOPA-quinone). This depletes the newly formed L-DOPA, and ascorbic acid or the like in the cell assists in the reduction of dopaquinone to dopa.

 

The second key enzyme is dopa dioxygenase, which catalyzes the formation of the intermediate 4,5-Seco-DOPA from L-DOPA.

 

The third key enzyme is cytochrome monooxygenase (Cyt P450), which catalyzes the formation of cyclic dopa (Cyclo-DOPA) [39].

 

Glucosyltransferase (UDP-G), the fourth key enzyme in the synthesis of beet pigments, is a modification enzyme that transfers the glycosyl group to beta in aglycone to form beta aglycone. Cyclic dopa forms cyclodopa glucose under the action of glucosyltransferase. Then it reacts spontaneously with betadine to form betanin, which is the basic aglycone formed by betalain. Another possibility is that cyclic dopa first reacts spontaneously with the chromophore betaaluronan to form betanin, and then forms betanin under the action of glucosyltransferase. Glucosyltransferase was isolated and purified from Dorotheanthus bellidiformis of the family Ankoaceae.

 

Application:

Currently, the European Union has approved betalains for use in food (EEC No. E162). The U.S. Food and Drug Administration (FDA) also officially approved the use of betalain in the food field (No. 73.40). Such pigments may be more widely used because they are more stable in high pH (>4) environments (their antioxidant capacity is 1.5 to 2 times that of anthocyanin pigments). Applications of beet coloring include candy, yogurt, salads, soft drinks, etc. Adding 50 mg·kg-1 beet pigment can have a good coloring effect. Phylloxanthin is endowed with unique physicochemical properties and health care value from betalain due to its special amino acid content. It will become an important source of supplementary amino acids in food.
