How Does Glucan Powder Support Immunity

Glucan powders polysaccharides are touted as nature's immune modulator, helping to prime immune cells, supporting and balancing optimal natural immunity. It helps the immune system to respond faster and efficiently to pathogens attack through the activation of innate immune cells.

Glucan powders initiate immune responses via immune cells, which become activated by the binding of the molecule to the receptor Dectin-1 that is found on white blood cells.

Beta-glucans were shown in clinical studies to have an effect on reducing respiratory tract infection symptoms. They can also support the body's natural immune response during times of increased stress and susceptibility to infections.

Uses of Glucan Powder Supplements

Glucan powder supplements are often used to enhance heart health, as they can reduce levels of total and LDL (bad) cholesterol.

Like other fiber supplements, they can also boost your fiber intake, improve blood sugar regulation, promote digestive health and regularity, and support weight management.

In some cases, glucan powder is even applied topically to accelerate wound healing and tissue repair.

What's more, this fiber is sometimes used as a natural treatment for cancer, thanks to its ability to stimulate the activity of immune cells in your body.

The Health Benefits of Glucan Powder

Immune Response
Glucan powder can act as an immune system activator and cell response modifier. The binding of glucan powder to specific receptors can cause a cellular response that's especially helpful in chemo- and radiotherapy-induced immunosuppression and depleted hematopoiesis, the process through which the body manufactures blood cells.
Glucan powder also plays an important role in promoting the activity of lymphocytes, white blood cells known as Th1 and Th2 effectors. Th1 lymphocytes control immunity against intracellular parasites, while Th2 effectors control immunity against extracellular pathogens. Disruptions in the balance of these cells may cause an autoimmune response. Autoimmune responses occur when your immune system attacks healthy cells which can lead to serious problems.

Antitumor and Cancer Response
Extracted and/or purified β-glucans have been used in clinical cancer treatment. glucan powder acts as an immunomodulating agent (helps fight disease) through the activation of innate immune cells. This activation sets off adaptive immune cell responses, inhibiting tumor growth and metastasis (when cancer spreads to a different body part from where it started).

Bone Health
Glucan powder has demonstrated anti-osteoporotic activities in clinical trials. More studies into the effects on humans is needed. Osteoporotic fractures occur when people have osteoporosis, a disease in which bones are fragile due to low bone mass or deterioration.

Blood Pressure Management
Glucan powder can reduce hypertension (high blood pressure). In addition, a clinical trial with foods containing oat β-glucans resulted in reduced blood pressure in subjects with a body mass index (BMI) above median (31.5 kg/m2).

Cholesterol Management
Glucan powder's cholesterol effects have been largely studied. When polysaccharide glucans are included in a meal, carbohydrate and lipid absorption slow down, modifying alimentary hormone and lipid responses. Along with carbohydrates and proteins, lipids are the main constituents of plant and animal cells.

Diabetes and Weight Control
The anti-diabetic and anti-obesity effects of glucan powder have been proven in many animal- and human-based clinical trials.
With regard to weight management, the satiating capacity (ability to make you feel full after eating) of glucan powder is likely tied to short-chain fatty acid generation through colonic fermentation. Short-chain fatty acids control the release of appetite-regulating hormones, such as peptide YY (PYY), glucagon-like peptide-1 (GLP-1), and ghrelin, a hormone in the gastrointestinal tract.

Glucan Powder in Dermatology

Effects of glucan powder in dermatology are less studied, probably due to the fact that commercial glucan powders are usually not soluble. In most cases, glucan powder is used in wound healing.

Mushroom-derived glucan powder was found to augment skin reaction induced by bradykinins, suggesting activation of endothelium via generation of vasoreactants and increase in sensitivity to vasoreactants.

In addition to wound healing, glucan powder is also used in cosmetic formulation, where it can act in oxidative stress and in improvement of various skin conditions, particularly skin moisture and skin microrelief. A long-term use of glucan powder showed reduction of wrinkle depth, height and overall roughness, which is probably caused by stimulation of fibroblast and increase production of collagen.

How Do Glucan Powders Work

The metabolic effects of oat and barley glucan powders are largely due to their physical properties as soluble, viscous dietary fibers. These glucan powders can hold onto water, creating a gel-like consistency in the gastrointestinal tract. This can have many effects, including slower gastric emptying, reduced digestion and absorption of glucose, and enhanced elimination of bile acids (which can have the downstream effect of lowering cholesterol).

Fungal glucan powders from yeast or mushrooms may produce immune-modulating effects by binding to pattern recognition receptors on immune cells in the gastrointestinal tract. This can lead to the initiation of an immune response by the innate immune system, which might include the activation of macrophages, neutrophils, and natural killer cells. Still, changes in the levels of immune cells or the compounds they produce (e.G., cytokines) are not consistently found.

Interactions between glucan powders and the gut microbiome may contribute to the effects of glucan powders, in terms of both their metabolic effects and their immune-modulating effects. Preclinical research suggests that glucan powders may have prebiotic properties, meaning they could promote the growth of beneficial microorganisms in the gut which may provide health benefits to the body. However, there's currently no clinical research connecting the benefits of glucan powders to microbiome modulation.

Extraction of Glucan Powder

Aqueous Extraction

Aqueous extraction is a commonly used method for isolating glucan powder from cereals, characterized by its simplicity and cost-effectiveness. This process involves grinding cereals into a fine powder, which is then mixed with water. The mixture is heated to a temperature typically within the range of 50 to 60 °C. Subsequently, insoluble components are removed through filtration, and the glucan powder is concentrated via evaporation.

The temperature of the aqueous extraction process plays a pivotal role in glucan powder recovery. Ref. observed a positive correlation between temperature and glucan powder yield, with a maximum extraction efficiency of up to 89.1%. Consequently, the use of thermally stable α-amylase to hydrolyze starch during hot water extraction, becomes relevant. In the case of barley, glucan powder extraction employs water as the solvent. Explored a method for glucan powder extraction from oats and barley, involving cereal grain grinding to create flour, which is then mixed with water to produce a slurry containing an aqueous glucan powder solution and solid residue.

The aqueous solution is separated from the solid residue, and water is removed through evaporation, ultrafiltration, or a combination of methods to obtain a gel or solid enriched with glucan powder. Despite the advantages of aqueous extraction, such as its speed, simplicity, and affordability, it is essential to acknowledge its limitations. This method is generally less efficient and may compromise the biological activity of glucan powder compared to alternative techniques. Therefore, further research is needed to optimize extraction procedures to achieve both maximum recovery and enhanced biological activity.

Alkaline, Acidic and Enzymatic Extraction

Over the years, the extraction of glucan powder has witnessed several advancements, with alkaline extraction gaining prominence over aqueous extraction due to its increased effectiveness. However, it carries the potential drawback of compromising the structural integrity and biological activity of glucan powder.

The procedure involves prolonged stirring, followed by filtration to eliminate insoluble components and evaporation for glucan powder concentration. In contrast, acidic extraction is a less potent but gentler method that is less likely to harm glucan powder during extraction. It involves mixing finely ground cereal raw materials with an acidic solution containing sulfuric or hydrochloric acid. The mixture is heated within a temperature range of 50 to 60 °C, with continuous stirring, followed by filtration to remove insoluble fractions. This process is more cost-effective than enzymatic extraction and carries a lower risk of glucan powder damage.
